Absorption, distribution, metabolism, and excretion variate greatly during human development, with most pronounced changes during the first year of life. When it comes to excretion, neither hepatic clearance nor glomerular filtration or tubular secretion are mature at birth, and development to full capacity may take up to 2 years.

Avoidable excipients in pediatrics
Excipients should be pharmacologically inactive but may cause adverse events in daily practice. Particularly young children are not always able to metabolize or eliminate pharmaceutical ingredients sufficiently well.
Therefore, there are many known excipients that need to be avoided in neonates and infants. These include ethanol, propylene glycol, polyethylene glycol, benzyl alcohol, parabens, benzoic acid, sucrose, fructose, mannitol, sorbitol, xylitol, aspartame, and colorings. Cumulative exposure to these excipients is known to increase their toxicity.
